Colorimetry is the science of measuring and describing colors, often using a colorimeter or spectrophotometer to quantify the color of a substance. Transmission refers to the passage of light through a substance, and is often measured as the ratio of transmitted light to incident light. Absorbance is a measure of how much light is absorbed by a substance, and is often used to quantify the concentration of a solute in a solution. Extinction is the reduction in the intensity of light as it passes through a medium, and is related to both absorption and scattering of light by the medium. **

Single-stranded DNA has a higher absorbance at 260 nm than double-stranded DNA because the bases in single-stranded DNA are more exposed to the surrounding environment, making them more accessible to absorb UV light. In contrast, the bases in double-stranded DNA are partially shielded by the complementary strand, reducing their exposure to UV light and thus lowering their absorbance at 260 nm. This difference in absorbance is commonly used in laboratory settings to quantify and characterize DNA samples. **
